Resumo: A elevada disponibilidade de energia na forma de triacilglicerol intramuscular (IMTG) está associada à produção de espécies reativas de oxigênio (EROS), estresse oxidativo, baixa atividade do ciclo do ácido tricarboxílico (CAT) e b-oxidação. Estes fatores, per se e em conjunto tem ação inibitória na cascata de sinalização da insulina no tecido muscular para a absorção da glicose. A este fenômeno, deu-se o nome de resistência à insulina, relacionada diretamente com o desenvolvimento da doença Diabetes Mellitus tipo II. Este trabalho, portanto, foi relacionar os fatores envolvidos com o desenvolvimento da resistência à insulina, e suas possíveis prevenções e tratamentos. Neste sentido, um restabelecimento na atividade normal mitocondrial através da expansão da atividade do CAT a custa de reações anapleróticas (entrada de substratos intermediários no CAT) poderia favorecer o consumo de substratos, incluindo os IMTG. Deste modo não haveria uma exposição crônica aos efeitos nocivos destas substâncias, melhorando a resposta da cascata de sinalização da insulina no tecido muscular. As contrações musculares contribuem neste processo, pois ativam a -oxidação dos ácidos graxos, além de possivelmente elevar o conteúdo de um poderoso antioxidante, a glutationa. Esta age contra o estresse oxidativo causado pelas EROS no tecido muscular, prevenindo o desenvolvimento da resistência à insulina. Assim como o exercício físico, o desacoplamento mitocondrial realiza papel importante na melhora do consumo de substratos. Desta forma, a proteína desacopladora mitocondrial (UCP-3) age na regulação da produção de EROs, protegendo as células musculares do estresse oxidativo, além de aumentarem o consumo de substratos. Todos estes processos contribuem na manutenção de uma resposta positiva na sinalização da insulina no músculo esquelético

Abstract: The high energy availability in form of intramuscular triglyceride (IMTG) is associated with the production of reactive oxygen species (EROS), oxidative stress, low activity of the tricarboxylic acid cycle (CAT) and b-oxidation. This factors, per se and in conjunct have an inhibitory action in the insulin pathway signalization in the muscle tissue for the absorption of glucose. This phenomenon is called of insulin resistance, and it is directly related to the developing of the Diabetes Mellitus type II disease. This work, therefore, was to link the factors concerned with the developing of the insulin resistance, and the possible preventions and treatment. Whereas, a recovery in the normal mitochondrial activity through the increase in the CAT flux, with anaplerotics reactions (entrance of intermediate substrates in the CAT) could enhance the consumption of substrates, including the IMTG. Thus there wouldn¿t be a chronic exposure to the harmful effects of these substances, improving the answer of the insulin pathway in muscle tissue. The muscle contractions contribute in this process, because they activate the -oxidation of the fat acids, besides it could possibly elevate the content of a powerful antioxidant, the glutathione. It acts against the oxidative stress caused by EROS in the muscle tissue, preventing the development of the insulin resistance. Similar to the exercise the mitochondrial uncoupling acts importantly in the improvement of the substrate consumption. Indeed the mitochondrial uncoupling protein (UCP-3) acts in the regulation of the EROS production, protecting the muscle cells of the oxidative stress, as well enhancing the substrate consumption.
