|Title:||KINETIC-PROPERTIES OF MAIZE GERM CYTOPLASMIC INORGANIC PYROPHOSPHATASE|
|Abstract:||An alkaline inorganic pyrophosphatase (pyrophosphate hydrolase EC 3.6. 1. 1.) was purified from maize (Zea mays) germ. This enzyme showed high specificty for inorganic pyrophosphate (PPi) with a K(m) value of 4.7 x 10(-5) M. The pH optimum was 8.0 to 8.5, in the presence of 5 mM Mg2+. The optimum Mg2+ : PPi ratio for maximum enzyme activity was 2.5. PPi or Mg2+ did not protect the enzyme against heat inactivation at 55-degrees-C. N-ethylmaleimide and dithio-bis-nitrobenzoic acid did not affect significantly the enzyme activity. An activation energy of 14.4 kcal mol-1 was determined for the reaction catalyzed by maize germ inorganic pyrophosphatase.|
