Abstract: Resumo: O presente trabalho descreve a síntese e caracterização química e espectroscópica do ligante 2-tiofen-2-il-imidazol (Thim) e de seus complexos inéditos de paládio(II) (Pd-Thim), platina(II) (Pt-Thim) e prata(I) (Ag-Thim). O ligante teve sua estrutura resolvida por primeira vez por difração de raio-X de pó. Os complexos Pd-Thim e Ag-Thim tiveram suas estruturas resolvidas por difração de raio-X de monocristal. Os complexos Pd-Thim e Pt-Thim possuem a fórmula mínima [PdCl2(Thim)2] e [PtCl2(Thim)2], sendo complexos neutros de geometria trans quadrado planar. O complexo Ag-Thim possui a fórmula mínima [Ag(Thim)2]NO3, sendo um complexo catiônico de geometria linear. O ligante se coordena aos metais pelo mesmo átomo em todos com complexos, o átomo de nitrogênio imino do grupo imidazol. Como não foi possível resolver a estrutura do complexo Pt-Thim comparações espectroscópicas com o complexo Pd-Thim e com o Thim foram efetuadas para se propor sua estrutura. Também foram realizados cálculos por DFT do Thim, Pd-Thim e Pt-Thim obtendo-se uma estrutura análoga ao Pd-Thim. Thim e Pd-Thim foram estudados quanto às suas interações com DNA de timo de bezerro por espectroscopia no UV-Vis, dicroísmo circular, fluorescência com brometo de etídio e determinação do ponto de desnaturação do DNA. Estas análises indicam que tanto o Thim quando o Pd-Thim interagem com o DNA e foi possível propor modos de interação para ambos os compostos. Estudos de atividade antibacteriana foram realizados para o Thim e os complexos e somente o Ag-Thim foi ativo contra as cepas testadas. Thim, Pd-Thim e Pt-Thim foram testados quanto à atividade antifúngica e somente o Pd-Thim se mostrou ativo. O Thim e os complexos foram testados quanto às suas citotoxicidades contra uma linhagem celular de câncer de mama e uma linhagem de osteoblasto. Todos os compostos apresentaram citotoxicidade contra as cepas testadas, sendo a mais expressiva para o Ag-Thim. O complexo Pd-Thim foi o único que mostrou seletividade frente às células tumorais

Abstract: This present work describes the synthesis and full chemical and spectroscopic characterization of the ligand 2-thiophen-2-yl-imidazole (Thim) and its novel complexes with palladium(II) (Pd-Thim), platinum(II) (Pt-Thim) and silver(I) (Ag-Thim). The ligand had its structure solved by powder X-ray diffraction. The Pd-Thim and Ag-Thim complexes had their structures solved by single crystal X-ray diffraction. The Pd-Thim and Pt-Thim complexes have the minimum formula [PdCl2(Thim)2] and [PtCl2(Thim)2], being neutral complexes with trans square plannar geometry. The Ag-Thim complex has the minimum formula [Ag(Thim)2]NO3, being a cationic complex with linear geometry. The ligand coordinates to the metals through the same atom in all the complexes, the imine nitrogen atom of the imidazole group. Spectroscopic comparisons were made between Pd-Thim, Pt-Thim and Thim to propose the Pt-Thim structure as it was not possible to solve it by X-ray diffracion. DFT calculations were also performed for Thim, Pd-Thim and Pt-Thim and the result was that the Pt-Thim structure is analogous to the Pd-Thim structure. Thim and Pd-Thim were evaluated for their interactions with calf-thymus DNA by UV-Vis spectroscopy, circular dichroism, fluorescence with ethidium bromide and denaturarion point of DNA. These analyses indicate that both Thim and Pd-Thim interact with DNA and it was possible to propose the modes of interaction. Antibacterial activity studies were performed for Thim and all the complexes, but only the Ag-Thim complex is active against the tested strains. Thim, Pd-Thim and Pt-Thim were evaluated for their antifungal activity, but only Pd-Thim is active. Thim and all the complexes were evaluated for their citotoxic activity agains a strain of breast cancer and one of osteoblast. All the compounds were citotoxic agains the testet strains with Ag-Thim being the most active. Pd-Thim was the only one that shows selectivity for the tumoral cells
